A conservative region of the mercuric reductase gene (merA) as a molecular marker of bacterial mercury resistance BJM
Sotero-Martins,Adriana; Jesus,Michele Silva de; Lacerda,Michele; Moreira,Josino Costa; Filgueiras,Ana Luzia Lauria; Barrocas,Paulo Rubens Guimarães.
The most common bacterial mercury resistance mechanism is based on the reduction of Hg(II) to Hg0, which is dependent of the mercuric reductase enzyme (MerA) activity. The use of a 431 bp fragment of a conservative region of the mercuric reductase (merA) gene was applied as a molecular marker of this mechanism, allowing the identification of mercury resistant bacterial strains.

A dual symbiosis shared by two mussel species, Bathymodiolus azoricus and Bathymodiolus puteoserpentis (Bivalvia : Mytilidae), from hydrothermal vents along the northern Mid-Atlantic Ridge ArchiMer
Duperron, Sebastien; Bergin, C; Zielinski, F; Blazejak, A; Pernthaler, A; Mckiness, Z; Dechaine, E; Cavanaugh, C; Dubilier, Nicole.
Bathymodiolus azoricus and Bathymodiolus puteoserpentis are symbiont-bearing mussels that dominate hydrothermal vent sites along the northern Mid-Atlantic Ridge (MAR). Both species live in symbiosis with two physiologically and phylogenetically distinct Gammaproteobacteria: a sulfur-oxidizing chemoautotroph and a methane-oxidizer. A detailed analysis of mussels collected from four MAR vent sites (Menez Gwen, Lucky Strike, Rainbow, and Logatchev) using comparative 16S rRNA sequence analysis and fluorescence in situ hybridization (FISH) showed that the two mussel species share highly similar to identical symbiont phylotypes. FISH observations of symbiont distribution and relative abundances showed no obvious differences between the two host species. In...

A laboratory model system for analysing microbial interactions occurring in anoxic estuarine sediments. ArchiMer
Herbert, R; Keith, S.
Defined mixed populations of Clostridium butyricum, Desulfovibrio desulfuricans and Chromatium vinosum co-existed for long periods when grown in a single stage chemostat with glucose as sole carbon source. The nature and availability of the inorganic nitrogen source exerted a major effect on carbon flow in the experimental system. The data show that both the C. butyricum and D. desulfuricans isolates can utilise NO sub(3) as an e super(-)acceptor with an increase in cell yield. Under these growth conditions the free S super(2-) levels were lower resulting in more stable mixed populations. In addition the data show that the C. butyricum produces more oxidised fermentation end-products (acetate) when NO sub(3) super(-) was available and more reduced products...

A microbial community analysis of the octocoral Eunicea fusca Electron. J. Biotechnol.
Duque-Alarcón,Angela; Santiago-Vázquez,Lory Z; Kerr,Russell G.
While there is a significant and growing body of knowledge describing the microbial communities of marine invertebrates such as sponges, there are very few such studies focused on octocorals. The octocoral Eunicea fusca is common on reefs in various regions of the Caribbean and has been the subject of natural product investigations. As part of an effort to describe the microbial community associated with octocorals, a culture-independent analysis of the bacterial community of E. fusca was conducted. Specifically, a 16S rDNA clone library analysis was performed to provide baseline data. A total of 40 bacteria members from 11 groups were found. In general, Proteobacteria were the dominant group with a total of 24 species and α-Proteobacteria...

A structural perspective on the mechanisms of quorum sensing activation in bacteria Anais da ABC (AABC)
LIXA,CAROLINA; MUJO,AMANDA; ANOBOM,CRISTIANE D.; PINHEIRO,ANDERSON S..
Bacteria are able to synchronize the population behavior in order to regulate gene expression through a cell-to-cell communication mechanism called quorum sensing. This phenomenon involves the production, detection and the response to extracellular signaling molecules named autoinducers, which directly or indirectly regulate gene expression in a cell density-dependent manner. Quorum sensing may control a wide range of biological processes in bacteria, such as bioluminescence, virulence factor production, biofilm formation and antibiotic resistance. The autoinducers are recognized by specific receptors that can either be membrane-bound histidine kinase receptors, which work by activating cognate cytoplasmic response regulators, or cytoplasmic receptors...

A test of community assembly rules using foliar endophytes from a tropical forest canopy ArchiMer
Donald, Julian; Roy, Mélanie; Suescun, Uxue; Iribar, Amaia; Manzi, Sophie; Péllissier, Léonie; Gaucher, Philippe; Chave, Jérôme.
1.Community assembly theory assumes that ecological communities are spatially delimited into patches. Within these patches, coexistence results from environmental filtering, competition, and immigration. Truly delineated communities exist in laboratory studies of microbial cultures in Petri dishes, yet empirical tests conducted in continuous environments often use patches defined by convention as opposed to realised boundaries. 2.Here we perform a test of ecological community assembly rules using foliar endophyte communities from a tropical rainforest, where leaves are considered as patches for both fungal and bacterial communities. We determined the diversity of fungal and bacterial endophytes using environmental DNA sequencing of 365 top‐canopy leaves,...

Adaptive features of a bacteriolytic enzyme from the style of the mussel Choromytilus meridionalis in response to environmental fluctuations. ArchiMer
Seiderer, L; Robb, F.
The extracellular digestive apparatus of bivalve molluscs consists of a crystalline style containing diverse degradative enzymes which are released during rotation and dissolution of the style. This paper reviews the occurrence of a temperature-regulated bacteriolytic enzyme in the black mussel Choromytilus meridionalis , which is an inhabitant of coastal reefs on the south and south west coast of South Africa. This agent is unable to lyse Micrococcus luteus or Escherichia coli , both of which are susceptible to lysis by a true lysozyme. It appears to have a restricted spectrum of activity which includes both free-living and kelp-associated gram-negative bacteria. In addition, the level of activity varies dramatically according to the environmental water...

An abyssal mobilome: Viruses, plasmids and vesicles from deep-sea hydrothermal vents ArchiMer
Lossouarn, Julien; Dupont, Samuel; Gorlas, Aurore; Mercier, Coraline; Bienvenu, Nadege; Marguet, Evelyne; Forterre, Patrick; Geslin, Claire.
Mobile genetic elements (MGEs) such as viruses, plasmids, vesicles, gene transfer agents (GTAs), transposons and transpovirions, which collectively represent the mobilome, interact with cellular organisms from all three domains of life, including those thriving in the most extreme environments. While efforts have been made to better understand deep-sea vent microbial ecology, our knowledge of the mobilome associated with prokaryotes inhabiting deep-sea hydrothermal vents remains limited. Here we focus on the abyssal mobilome by reviewing accumulating data on viruses, plasmids and vesicles associated with thermophilic and hyperthermophilic Bacteria and Archaea present in deep-sea hydrothermal vents.

Analyse bacteriologique de l'eau par mesure potentiometrique de la reduction de l'acide lipoique ArchiMer
Charriere, G; Jouenne, T; Junter, G.
A simple potentiometric technique to detect and enumerate the bio-indicators of water contamination is described. The technique consists in measuring the redox potential of a specific culture medium saturated with oxygen where bacteria are incubated in presence of lipoic acid (LA). These micro-organisms reduce LA to dihydrolipoic acid, giving rise to an important potential drop in the system. This potentiometric "signal" reveals the presence of active metabolic bacteria in the medium. The major assets of potentiometric measurements in presence of LA lies in the reliability and intelligibility of the results (the redox phenomenon being qualitatively and quantitatively studied), associated to a very simple technology. Futhermore, this technique is perfectly...

Antarctic Psychrophile Bacteria Screening for Oil Degradation and Their Degrading Characteristics. OceanDocs
Zheng, Zhou; Jin, Qing; Miao, Jinlai; Liu, Fangming.
Two hydrocarbon degrading bacteria NJ276 and NJ341 were screened from 385 Antarctic marine bacteria and their degrading characteristics were studied. Diesel oil as sole carbon source was used in this study. The results showed that the oil degradation rates of Antarctic psychrophile bacteria NJ276 and NJ341 were 23.47 % and 32.15 %, respectively, after 20 days culturation at 5 °C, and the rates were 43.95 % and 62.47 % respectively after 20 days culturation at 15 °C. The oil degradation abilities were enhanced remarkably with the increasing culture temperature. GC - MSs indicated the residual oil contained C15 - C21 7 alkyls after degradation by NJ276, and C16, C17 and C18 3 alkyls after degradation by NJ341. The 16S rDNA gene sequences homology and...

Antibacterial activity of different types of snake venom from the Viperidae family against Staphylococcus aureus Biological Sciences
Canhas, Isabela Nascimento; Heneine, Luiz Guilherme Dias; Fraga, Thaís; Assis, Débora Cristina Sampaio de; Borges, Márcia Helena; Chartone-Souza, Edmar; Nascimento, Andréa Maria Amaral.
Toxins and venoms produced by living organisms have exhibited a variety of biological activities against microorganisms. In this study, we tested seven snake venoms from the family Viperidae for antibacterial activity and the activities of reversal of antibiotic resistance and inhibition of biofilm formation against 22 clinical isolates of Staphylococcus aureus. Bothrops moojeni venom exhibited anti staphylococcal activity with the lowest mean value of minimum inhibitory concentration (MIC). Moreover, reversal of antibiotic resistance was observed for combinations of B. moojeni venom (½ x MIC) and norfloxacin or ampicillin (both ½ x MIC) for 86.4% and 50% of the isolates, respectively. B. moojeni venom alone at ½ MIC inhibited 90% of biofilm formation,...

Antifouling effect of bioactive compounds from marine sponge Acanthella elongata and different species of bacterial film on larval attachment of Balanus amphitrite (cirripedia, crustacea) BABT
Ganapiriya,Viswambaran; Maharajan,Athisuyambulingam; Kumarasamy,Pothiappan.
The antifouling activity of bioactive compounds from marine sponge Acanthella elongata (Dendy) and five species of bacterial biofilm were studied. Larvae of Balanus amphitrite (Cyprids and nauplii) were used to monitor the settlement inhibition and the extent to which inhibition was due to toxicity. The crude extract and partially purified fractions of A.elongata showed significant inhibition over the settlement individually, and with the interaction of bacterial species. No bacterial film stimulated the barnacle settlement. The high but variable levels of antifouling activity in combination with less amount of toxicity showed the potential of these metabolites in environmentally-friendly antifouling preparations.
